PDCD1 分子靶点综述

CD279,又名程序性细胞死亡蛋白1(PD-1),由PDCD1基因编码。PD1为免疫球蛋白超家族成员,与CD28、CTLA-4和ICOS分子同属于CD28超家族,主要以单体形式表达于活化的CD4+T和CD8+T细胞及B细胞,其配体B7-H1和B7-DC。研究证实,增强PD-1的信号可以下调已活化的T、B细胞在外周组织中的应答,在维持外周T、B细胞耐受中发挥重要的作用。

Located in external side of plasma membrane. Is expressed in retina. Used to study dilated cardiomyopathy and systemic lupus erythematosus. Human ortholog(s) of this gene implicated in autoimmune disease (multiple); hepatitis B; hepatitis C; hepatocellular carcinoma; and lupus nephritis. Orthologous to human PDCD1 (programmed cell death 1).

操作步骤

使用方法

全血样本:取 100 μl 抗凝血,加入 5 μl Anti-Mouse CD279, APC 和其他抗体,震荡混匀,于室温避光孵育 15 分钟或 2 - 8℃避光孵育 30 分钟。根据红细胞裂解液说明书进行操作,去除红细胞。300× g 离心 5 - 10 分钟,弃上清,加入 500 μl 流式染色缓冲液重悬,上机检测。
其他样本:将 1×105 - 108 个单细胞经适当离心后,取适量流式染色缓冲液 (目录号 S1001)重悬,加入 5 μl Anti-Mouse CD279, APC 和其他抗体,使反应终体积为 100 μl。震荡混匀,于室温避光孵育15 分钟或 2 - 8℃避光孵育 30 分钟。每管加入 1 - 2 ml 流式染色缓冲液,300 × g 离心 5 - 10分钟,弃上清,加入 500 μl 流式染色缓冲液重悬,上机检测。
实验中,可对抗体进行优化滴定以获得最佳效果。
