检测原理:本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性捕获抗体预包被在高亲和力的酶标板上,酶标板孔中预先加入生物素标记的检测抗体微球和辣根过氧化物酶微球。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的待测物质与捕获抗体和检测抗体结合。洗涤去除未结合的物质后,加入显色底物(TMB),避光显色。颜色反应的深浅与样本中待测物质的浓度成正比。加入终止液终止反应,在 450 nm 波长 (参考波长 5630 nm) 测定吸光度值。

IL10 分子靶点综述

白细胞介素10(IL-10)是由2个长178个氨基酸的亚基构成的同源二聚体。在人类中,它由位于1号染色体的IL-10基因编码,由5个外显子组成。IL-10主要由单核细胞分泌,淋巴细胞、II型辅助性T细胞(Th2)、肥大细胞、CD4+CD25+Foxp3+调节性T细胞和部分活化的T细胞和B细胞分泌较少。它对免疫调控和炎症具有多种效应。它下调巨噬细胞的Th1细胞因子、II类MHC抗原和共刺激分子。它还能增强B细胞存活、增殖和抗体分泌。IL-10能阻断NF-κB活性,并参与JAK-STAT信号通路调控。IL-10的免疫抑制特性提示其可能应用于临床,抑制器官移植后的排斥。IL-10还具有强烈的消炎特性。

Located in extracellular space. Is expressed in colon; liver; thymus; and thymus primordium. Used to study inflammatory bowel disease. Human ortholog(s) of this gene implicated in several diseases, including artery disease (multiple); autoimmune disease (multiple); eye disease (multiple); gastrointestinal system cancer (multiple); and hematologic cancer (multiple). Orthologous to human IL10 (interleukin 10).
