检测原理：本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠Fas抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体，经过孵育，样本中存在的Fas与固相抗体和检测抗体结合。洗涤去除未结合的物质后，加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后，加入显色底物TMB，避光显色。颜色反应的深浅与样本中Fas的浓度成正比。加入终止液终止反应，在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

FAS 分子靶点综述

Fas，又名APO-1、CD95或TNFRSF6，是一个可转导凋亡信号的细胞表面受体。它是两种凋亡途径之一，另一种是线粒体途径。一旦结合配体，Fas就会形成死亡诱导信号复合物。相邻细胞表面的膜锚定Fas配体三聚体会导致Fas低聚化。近来的小鼠模型研究表明，Fas促进其肿瘤生长，但是人类癌症基因组学数据库表明Fas可作为肿瘤抑制物。在AOM-DSS诱导的结肠癌和MCA诱导的肉瘤小鼠模型中，结果显示Fas是肿瘤抑制物。另外，Fas还可介导肿瘤特异性细胞毒性T细胞的抗肿瘤细胞毒作用。研究报道，癌症和自身免疫性疾病患者的血清Fas水平是升高的，前者可能来源于肿瘤细胞本身。

Acts upstream of or within several processes, including extrinsic apoptotic signaling pathway; hematopoietic or lymphoid organ development; and regulation of hemopoiesis. Located in external side of plasma membrane and extracellular region. Part of CD95 death-inducing signaling complex. Is expressed in several structures, including alimentary system; brain; genitourinary system; hemolymphoid system gland; and liver and biliary system. Used to study Sjogren's syndrome; autoimmune lymphoproliferative syndrome; systemic lupus erythematosus; and type 1 diabetes mellitus.
